The latest death toll reported by the department on Friday stood at 578.
The new spike in cases comes as people in Northern Ireland were advised not to travel across the border to the Republic unless it is “absolutely necessary”.
It is understood that border checks will be carried out during Saturday in response to the surging cases in Donegal.
